Same code, Same IIS version- different results

So yes, I know something is different- just not sure what. I have been tasked with moving the application from a very old server to a new server.  I was told it was tried before but there was an issue with moving it.

I moved it to my local machine, set the proper configs, and got it running

1: local machine (Win 7 64bit)  IIS 7.5.7600.16385 
2: Test machine ( Win Server 2008R2 64bit)  IIS 7.5.7600.16385

I copied the sub-directory from Machine 1 to Machine 2, Configured the new Site, Put it in a new application pool and configured it identically (2.0, Classic mode, 32 bit allowed)
Authentication on both are Windows (all others disabled) and Negotiate security
ASP.NET 2.0 is properly registered with the site
Handlers are correct.

On the local machine everything works as expected.
The server I get the "Extender controls may not be registered after PreRender." error.

Eveything I have seen in this forum as well as elsewhere on the internet, as expected, point towards a control being registered and offer ways to re-code to work that out.

1. I am trying to determine how it's code, if it's identical
2. I did not write the application, and I don't know the AJAX Control Toolkit.
3. I don't have the original project/code.

I think it seems to me that this has to be something config related- but I have seen no answers anywhere that point to a config issue.

Has anyone run into something similar? How have you resolved?
